L-Azidonorleucine (ANL) is a noncanonical, slow to metabolize, and clickable amino acid that can be incorporated to proteins in cells that express a mutant methionyl-tRNA synthetase (MetRS). The newly synthesized MetRS-expressing cell-derived proteins from cell mixtures can then be enriched and examined in a cell-type-specific manner. The azido handle in ANL can be selectively conjugated to dyes and functionalized beads to enable visualization of proteins by FUNCAT (Fluorescence non-canonical amino acid tagging), and to purify proteins using BONCAT (bio-orthogonal non-canonical amino acid tagging).
