Biochem/physiol Actions

Hydroxyphenyl-fluorescein (HPF), a cell-permeable and photostable fluorescent probe, is specifically designed for the selective detection of primarily hydroxyl radicals (HO•), and peroxynitrite (ONOOˉ) over minimally reactive H2O2, O2ˉ•, NO, 1O2 and ROO•. Initially exhibiting weak fluorescence (ε454 = 28,000 M-1cm-1, Φ: 0.006), HPF undergoes O-dearylation upon reaction with these species, releasing its hydroxy-phenoxy group and generating intensely fluorescent fluorescein, which leads to a dose-dependent fluorescence signal increase (λex/λem: 490/515 nm) and significantly enhanced optical properties (ε492 = 84,000 M-1cm-1, Φ: 0.85). HPF′s inherent selectivity, complete autoxidation resistance, low toxicity, and quantitative fluorescence response establish it as a valuable tool for applications spanning microscopy, enzymatic assays, and multiplexing.
