Isoquercetin (Quercetin-3-O-glucoside) functions as a highly orally bioavailable "pro-drug" for quercetin, leveraging SGLT1-mediated active transport for rapid intestinal absorption. Following uptake, it undergoes immediate deglycosylation and extensive phase II metabolism—including glucuronidation, sulfation, and methylation—within enterocytes and the liver, circulating primarily as conjugated metabolites. Mechanistically, Isoquercetin delivers potent pleiotropic efficacy: it inhibits oncogenic Wnt/β-catenin signaling, blocks viral replication via SARS-CoV-2 3CLpro and Influenza A PB2 (ED50 1.2 µM) inhibition, and exerts significant antithrombotic activity by inhibiting Protein Disulfide Isomerase (PDI; IC50 2.5 μM) and blocking platelet factor Va/thrombin generation (IC50 ~5 μM). Additionally, it restores redox homeostasis through Nrf2/HO-1 induction and suppresses neuroinflammation via NF-κB downregulation and BDNF upregulation.
