3-Azido-7-hydroxycoumarin (AHC) is a cell-permeable, pro-fluorogenic probe engineered for high-contrast bioorthogonal labeling. Initially non-fluorescent, AHC undergoes a highly specific click reaction with terminal alkynes to form a stable triazole product, eliciting a potent, "turn-on" blue-green fluorescence (λex/λem: ~403/478 nm) with a quantum yield of 0.79. This mechanism provides an exceptional signal-to-noise ratio, with fluorescence enhancement often exceeding 100-fold. Due to its high reaction efficiency under mild conditions, this versatile tool enables applications from protein labeling to the rapid detection of DNA in proliferating cells, activating its photostable signal only upon reacting with a specific DNA analog. Its primary strategic advantage is the elimination of background, which simplifies workflows and facilitates near real-time visualization of molecular dynamics, making it ideal for high-throughput screening.
