Biochem/physiol Actions

DPhPC (1,2-Diphytanoyl-sn-glycero-3-phosphocholine) is an amphiphile that self-assembles into exceptionally stable model bilayers. Structurally defined by two branched phytanoyl chains (tetramethylated 16:0 fatty acids) at the sn-1 and sn-2 positions, its bulky hydrophobic tails create significant steric hindrance. This architecture prevents the formation of a rigid crystalline phase, resulting in a larger area per lipid (~81.2 Ų) and eliminating the gel phase transition. Consequently, the membrane remains in a consistent liquid-crystalline state, providing high resistance to oxidation and mechanical stress. This stability, combined with its zwitterionic phosphocholine headgroup that influences the hydration layer and electrostatic interface, makes DPhPC an ideal matrix for reconstituting ion channels, studying protein-lipid interactions, and developing robust nanoliposomes.
