General description
The ZytoLight SPEC FGFR1 Dual Color Break Apart Probe (PL124) is a direct-labeled FISH probe designed for the qualitative detection of translocations involving the FGFR1 gene at 8p11.22-p11.23 in cytologic specimens, such as leukemic cells. It is intended for use with the ZytoLight FISH-Cytology Implementation Kit.
ZytoLight® SPEC FGFR1 Dual Color Break Apart Probe:
FISH uses direct-labeled probes flanking FGFR1 breakpoint that co-denature with target DNA in pretreated cytologic specimens (10 µl/slide, sealed), hybridize overnight (72°C/5 min denaturation, 37°C hybridization), then undergo stringent washing and DAPI counterstaining for visualization via fluorescence microscopy per ZytoLight FISH-Cytology Implementation Kit (Z-2099-20).
ZytoLight® SPEC FGFR1 Dual Color Break Apart Probe:
- ZyGreen labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting sequences proximal to the FGFR1 breakpoint region on -chromosome 8 (8p11.22).
- ZyOrange labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences distal to the FGFR1 breakpoint region.
- Formamide-based hybridization buffer.

Application
ZytoLight® SPEC FGFR1 Dual Color Break Apart Probe assists in identifying FGFR1 translocations in cytologic samples, within the context of the prescribed FISH workflow and in conjunction with the specified kit.
Analysis Note
In a normal interphase nucleus without translocations, two orange/green fusion signals are expected. In cells with FGFR1 translocations, an aberrant signal pattern may consist of one orange/green fusion signal, one orange signal, and one separate green signal, indicating the presence of a rearrangement.
