ZytoLight^®SPEC MCL1/1p12 Dual Color Probe

The ZytoLight SPEC MCL1/1p12 Dual Color Probe (PL129) is a direct-labeled FISH probe intended for the qualitative detection of amplifications involving the human MCL1 gene and for detecting specific sequences on chromosome 1p12 in formalin-fixed, paraffin-embedded (FFPE) specimens. It is designed to be used in conjunction with the ZytoLight FISH-Tissue Implementation Kit.

ZytoLight®SPEC MCL1/1p12 Dual Color Probe comprises:

• ZyGreen labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting the MCL1 gene region on chromosome 1 (1q21.3).
• ZyOrange labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences specific to chromosome 1p12.
• Formamide-based hybridization buffer.

Principle:
FISH detects specific sequences via direct-labeled probes co-denatured (75°C/10 min) with target DNA in pretreated FFPE sections (10 µl/slide), hybridized overnight (37°C), stringency washed, DAPI counterstained, and visualized by fluorescence microscopy per ZytoLight FISH-Tissue Implementation Kit (Z-2028-5/-20).

For the qualitative detection of human MCL1 gene amplifications and chromosome 1p12 specific sequences by fluorescence in situ hybridization (FISH)

In a normal interphase nucleus, two orange and two green signals are expected. In cases of MCL1 amplification, multiple green signals or clusters will be observed. Unexpected signal patterns should be investigated further for potential genetic rearrangements.
