General description
DNA topoisomerase 2-beta (UniProt: Q02880; also known as DNA topoisomerase II, beta isozyme, EC:5.6.2.2) is encoded by the TOP2B gene (Gene ID: 7155) in human. The TOP2B gene is located on human chromosome location 3p24.2. Topoisomerase IIβ (TOP2B) is an isoform of type IIA topoisomerases. It is a member of the topoisomerase II family and is a Key decatenating enzyme that alters DNA topology by binding to two double-stranded DNA molecules, generating a double-stranded break in one of the strands, passing the intact strand through the broken strand, and religating the broken strand. It also plays a role in B-cell differentiation. Structurally, TOP2B consists of an N-terminal domain that facilitates DNA binding and a C-terminal domain that harbors ATPase activity, enabling the enzyme to introduce double-strand breaks in DNA to alleviate torsional strain. TOP2B is predominantly expressed in various tissues, including the brain, heart, and skeletal muscle, with notable localization in the nucleus, where it performs its role in DNA manipulation. Two isoforms of TOP2B have been described that are produced by alternative splicing. TOP2B has been implicated in neurodegenerative disorders, as its dysfunction can disrupt neuronal homeostasis. Its involvement in disease is particularly significant in cancer, where aberrant expression or mutations in TOP2B can lead to genomic instability and contribute to tumorigenesis. Recent studies have highlighted the potential of TOP2B as a therapeutic target, particularly in the context of chemotherapy, where its inhibitors are used to enhance the efficacy of anticancer agents. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application
Evaluated by Western Blotting in SH-SY5Y cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TOP2B in SH-SY5Y cell lysate.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected TOP2B in human kidney tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound TOP2B peptide with at least one billion-fold (1,000,000,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected TOP2B in NIH/3T3 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
