General description

Las lamininas son glucoproteínas adhesivas no colagenosas[1] con un peso molecular sustancial. Son proteínas heterotrímeras con tres polipéptidos unidos por puentes de disulfuro conocidos como cadenas α, β y γ.[2] Los múltiples dominios polipeptídicos de las lamininas pueden unirse a varios receptores de la superficie celular, como diversos dímeros de integrina, distroglucano y sindecanos, y a otros componentes de la matriz extracelular (ECM). Esta glucoproteína trímera multidominio de la matriz extracelular es el principal componente de la lámina basal que propicia la adhesión, la proliferación y la diferenciación.

Application

La laminina de la membrana basal del sarcoma murino Engelbreth-Holm-Swarm se ha utilizado en:
  • la diferenciación de las células productoras de insulina (IPC).[3]
  • Ensayos de inhibición de la infección.[4]
  • Ensayos de proliferación .[5]
La laminina soporta el crecimiento y la diferenciación de muchos tipos de células, como las células epiteliales, endoteliales, neuronales, musculares y hepáticas. Se recomienda su uso como substrato de cultivo celular a 1-2 μg/cm2. La concentración óptima depende del tipo de célula, así como de la aplicación o de los objetivos experimentales.

Biochem/physiol Actions

Las proteínas de la laminina son componentes integrales del andamiaje estructural de los tejidos animales. Se asocian con el colágeno de tipo IV a través de entactina y perlecán y se unen a las membranas celulares a través de receptores de integrina, complejos de glucoproteínas y distroglucanos y glucoproteínas del grupo sanguíneo luterano. La laminina tiene dominios activos para la unión del colágeno, la adhesión celular, la unión de la heparina y el fragmento de crecimiento de las neuronas.

Preparation Note

Este producto se suministra a una concentración de 1 − 2 mg/ml en Tris-HCl 50 mM, con NaCl 150 mM. Se filtra utilizando un filtro de 0,2μm. Descongele lentamente esta disolución antes de utilizarla a 2-8°C para evitar la formación de gel. Para utilizar como recubrimiento, dilúyala en una disolución salina equilibrada, recubra la superficie de cultivo con un volumen mínimo e incube a 37 °C durante dos horas. Lave 3 veces con PBS antes de sembrar las células. Los revestimientos de laminina pueden almacenarse durante un mes a 2-8°C.

Other Notes

La laminina es una glucoproteína trimérica multidominio de matriz extracelular y es el principal componente no colágeno de la lámina basal que apoya la adhesión, la proliferación y la diferenciación. La laminina está compuesta por cadenas A, B1 y B2, que están conectadas por muchos enlaces de disulfuro. Esta laminina se aisló del tumor Engelbreth-Holm-Swarm de ratón.

    1. In undiluted form or diluted to working concentration, this material may be stored at -20°C in aliquots up to the recommended retest date for the specific lot. This data is published in the Certificate of Analysis. Avoid repeated freeze/thaw cycles. The solution may be transferred to smaller vials in working volumes for long-term storage.   4. What type of laminin is Product L2020?

    1. In the nomenclature system by Burgeson, this is Laminin 1. The laminins were numbered with Arabic numerals in the order discovered. The previous A, B1, and B2 chains, and their isoforms are listed by the Greek letters, for alpha, beta, and gamma, respectively, followed by an Arabic numeral to identify the isoforms.  For example, the first laminin identified from the Engelbreth-Holm-Swarm tumor (EHS) was designated laminin-1 with the chain composition α-1/β-1/γ-1. The genes for these 3 chains are LAMA1, LAMB1, and LAMC1. Reference: Burgeson, R.E., et al., A new nomenclature for the laminins. Matrix Biol., 14, 209-211 (1994).

  7. Can I coat Laminin, Product L2020, on top of poly-L-lysine for better adherence?

    1. When coating coverslips or slides for use with neural stem cells (neurospheres), the following procedure is recommended. Precoat the coverslips with poly-L-lysine (Product No. P5899) at 100 μg/mL. This is incubated for 30 minutes or more at room temperature. The coverslips are then washed with sterile water and allowed to dry. The poly-lysine coated coverslips can be stored at room temperature until ready to use (up to ~1 month). Coverslips are then coated with 30 - 50 μL of warmed laminin, which is at a concentration of 50 μg/mL. Incubate at 37°C for 30 minutes. Remove the laminin, and wash with DME (50 μL) twice. Cells are then added to the coverslips.

  8. How can I store solutions of Laminin, Product L2020?

    1. The product has a recommended storage temperature of -20°C.  Please note, the product needs to be thawed slowly at 2-8°C.  If the product has been warmed up at room temperature and it gels, it cannot be reactivated for use. We recommend storage at 2-8°C for up to one week. For longer storage, the product needs to be frozen at -20°C in single-use aliquots.
