General description

El medio RPMI 1640 lo desarrolló el equipo de Moore en el Instituto Roswell Park Memorial en 1966. Se formuló una modificación del medio McCoy 5A para sostener las células linfoblastoides en cultivo en suspensión, pero desde entonces se ha demostrado que sirve de soporte para una amplia variedad de células que necesitan anclaje. Originalmente indicado para utilizar con un complemento sérico, se ha demostrado que el RPMI 1640 respalda diversas líneas celulares en ausencia de suero. También se ha utilizado ampliamente en protocolos de fusión y en el crecimiento de células híbridas. Este medio es adecuado para el cultivo de leucocitos humanos sanos y neoplásicos.

Application

El Medio RPMI-1640 se ha utilizado para el mantenimiento de líneas celulares y actúa como medio para diferentes células.[1][2]

  1. When the medium waas sterile-filtered, which pore size was used?

    1. Endotoxin-free products are all typically filtered with a 0.1 micron pore size filter.  Sterile filtered products typically have a 0.2 or 0.22 pore size filter.

  2. Good Morning, does RPMI medium R8758 which contains sodium bicarbonate, affect cell suspension culture of megakaryoblastic cell line DAMI? I mean, sodium bicarbonate can significantly modify cell behavior or his effects can be tolerated?

    1. This product has not been validated for use with the megakaryoblastic cell line DAMI. However, literature indicates compatibility with this cell line. The following citation may be of interest: PLoS One. 2020 Apr 7;15(4):e0230507.

  3. I bought this product and realized that it does not contain Hepes. Can I add hepes?

    1. This product utilizes a bicarbonate buffering system. However, there are no anticipated problems with the addition of HEPES. We do offer RPMI formulations containing both sodium bicarbonate and HEPES, such as Products R5886 & R7638.

  4. Considering that I have to use an RPMI medium containing 4mM of l-Glutamine and 10mM of non-essential amino acids, can I use the product R8748 without adding anything?

    1. We do not recommend freezing cell culture media. A precipitate may form upon thawing.  Also, this can cause a loss of amino acids, which may yield low cell growth.

    1. There is no HEPES in this formulation of RPMI.  We do offer RPMI containing HEPES as Product No's. R5886 and R4130. The formulation of RPMI and other classical media can be found on the Cell Culture Web Page.
