The 91A kit is formulated for 10 uses. The Instructions For Use direct the user to prepare 48 mL of staining solution, which is enough to fill a standard Coplin jar. It is possible to stain just 1 slide per run, or up to 10 slides per run for a standard Coplin jar that holds 5 slides, if the slides are placed back to back. The number of slides that can be stained should fall between 10 slides per kit to 100 slides per kit. If the Coplin jar holds more than 5 slides, the number of slides stained per kit would be two times the number of slides the Coplin jar can hold.

Application
Esterasen sind in der Natur ubiquitär vorhanden; zu ihnen gehört eine Reihe unterschiedlicher Enzyme, die jeweils spezifische Substrate umsetzen. Gemäß der Methodik wird α-Naphthylacetat enzymatisch hydrolysiert, wobei eine freie Naphtholverbindung freigesetzt wird. Diese bindet anschließend an eine Diazoniumverbindung, wodurch an Orten nicht-spezifischer Esteraseaktivität schwarze Ablagerungen entstehen. Dieses Enzym wird hauptsächlich in Monozyten, Makrophagen und Histozyten nachgewiesen und ist normalerweise in Granulozyten nicht vorhanden. Lymphozyten können gelegentlich Enzymaktivität an den Tag legen. Ein Verfahren zur Natriumfluorid-Hemmung kann ausgeführt werden, um die Inhibierung von Monozyten zu belegen. Das Verfahren wird bei 37 °C an Blut, Knochenmarkfilmen, Touch-Gewebepräparaten, Zytozentrifugen-Präparaten und Gefrierschnitten durchgeführt. Das Enzym wird durch eine Paraffin-Aufbereitung abgetötet.
Für den zytologischen Nachweis von α-Naphthylacetatesterase im Blut, in Knochenmarkfilmen oder Touch-Gewebepräparaten vorgesehen.
