Application

Hyaluronidase from sheep testes has been used:
  • in flow cytometry[1]
  • in the isolation of putative spermatogonial stem cells (SSCs)[2]
  • to study the effects of magnetic stimulation in magnetic hydrogel properties[3]
  • in immunohistochemistry to visualize collagen type II distribution[4][5]
  • to expose group-III auditory neurons of Muller′s organ for patch-clamp recordings[6]
  • for the degeneration of hydrogels[7]

Biochem/physiol Actions

These enzymes randomly cleave β-N-acetylhexosamine-(1−4)-glycosidic bonds in hyaluronic acid, chondroitin, and chondroitin sulfates.

General description

Hyaluronidase is a hyaluronic acid-metabolizing enzyme.[8] It is found in the venoms of snake, honeybee and hymenoptera. Hyaluronidase degrades hyaluronic acid (HA), which enhances membrane permeability, lowers viscosity and increases tissue permeability. It enhances the diffusion of local anesthetics, lowers the size of myocardial infarction, improves the effects of anticancer drugs and prevents perineal trauma during spontaneous delivery.[9]

Other Notes

One unit will degrade 0.75 μg of the polysaccharide hyaluronic acid per minute at pH 5.35 at 37 ºC (as measured by turbidimetric absorbance (λ: 600nm) when complexed with BSA after 45 minutes).

Uncover more about glycosaminoglycans and proteoglycans including the structure of glycosaminoglycans (GAGs), the different types of GAGs, and their functions.

Glycosaminoglycans are large linear polysaccharides constructed of repeating disaccharide units.

Protocolos

To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.

Preguntas

1–2 de 2 Preguntas
  1. Kann die Hyaluronidase H2126 in PBS gelöst werden um sie dann in kleinen Aliquoten bei -20 C zu lagern?

    1 respuesta
    1. The lyophilized powder may be reconstituted at 5 mg/mL in 0.02M sodium phoshate buffer or 0.45% sodium chloride containing 0.01% BSA. Solutions prepared at concentrations of at least 1 mg/ml may be stored in aliquots at -20°C for several months.

  2. H2126-100MG Hyaluronidase from sheep testes についてですが、溶解方法を教えてください。

    1 respuesta
    1. This enzyme is soluble at 1 mg/mL in 20 mM sodium phosphate buffer (pH 7.0), with 77 mM sodium chloride and 0.1 mg/mL BSA at 1 mg/mL. For assay purposes, this product is prepared at 0.3 mg/mL solution in Phosphate Buffer. Heating may be required, Please see the link below to review the full assay protocol:
