General description
The presence of brilliant green, ox bile, high concentrations of thiosulfate and citrate largely inhibits the accompanying flora and swarming of Proteus spp, while allowing the growth of Salmonella spp. The synergistic reaction between thiosulfate and iron salt present in the media enables the growth of H2S positive black color colonies.
Lactose present in the media differentiates between the lactose positive and lactose negative microorganisms. Lactose fermenting microbes form an acid which is indicated by forming red colonies due to the presence of a pH indicator, neutral red. Peptones provide nitrogen, vitamins, amino acids, and minerals for growth and agar acts as a solidifying agent.
Application
Analysis Note
- Salmonella
- spp. produce translucent colonies with or without black center and yellowish-brown colored medium or may appear as almost black colonies due to H2S production.
- Shigella spp. produce translucent colonies without black center and yellowish-brown colored medium.
