Application

Probenecid has been used:
  • as a component of phenol red free Hank′s buffer for Intracellular Ca2+-mobilization assay[1]
  • as a component of calcium sensitive dye for calcium mobilization Studies[2]
  • as a supplement in Hank′s balanced salt solution (HBSS) assay buffer[3]

Useful as an inhibitor of the organic anion transporter, MRP.

Biochem/physiol Actions

Probenecid is a uricosuric agent, which increases uric acid removal in the urine[4] and reduces urate reuptake.[5] It also reduces the renal tubular excretion of many other drugs and increases their plasma concentration. The inhibitory action of probenecid is mediated by organic anion transporters. It is found useful for the treatment of gout.[5] Probenecid is responsible for the rise in the levels of neurochemicals in the brain by preventing its movement from choroid plexus and parenchyma cells into plasma.[4]

Questions

  1. What is the solubility and solution stability?

    1 answer
    1. The solubility specification was tested at 100 mg/ml in 1 M NaOH (refer to the specification sheet). Probenecid is soluble in chloroform, in dilute solutions of sodium hydroxide, then buffered to pH 7.4, and is nearly insoluble in water. It is also soluble in 1 M NaOH (50 mg/ml), producing a clear, colorless solution.
