Application

Mineral oil has been used as an overlay in microdrop embryo cultures.

Analysis Note

This material is tested as an overlay in microdrop embryo cultures. (All embryo tested materials are tested using mouse embryos.) Typically BCF1 x BDF1 hybrids are used. The oil is primarily used to prevent evaporation of the medium, particiularly when performing embryo manipulations.1 The procedure is as follows: 1-cell (BCF1 X BDF1) embryos were cultured in triplicate microdrops of embryo-tested HTF (Human Tubal Fluid) medium supplemented with 0.4% BSA. For a valid assay, at least 70% of 1-cell stage control embryos must develop to the blastocyst stage within 96 hours. For product release, at least 80% of 1-cell stage test embryos must develop to the blastocyst stage within 96 hours with the tested material.

Disclaimer

This product is not sterile. Mineral oil is difficult to sterilize. It can only be sterilized by careful filtration of the warmed oil. It is assumed antibiotics will be added to the cell culture medium. Otherwise, Prod. No. M5310 should be used, as it is sterile filtered.

Questions

1–7 of 7 Questions
  1. Is this product suitable for mineral oil analysis in water on FTIR ?

    1 answer
    1. This product is a light mineral oil (neat) intended for laboratory applications such as cell culture overlay and is not designed or validated as an analytical standard. FTIR can analyze mineral oils due to characteristic hydrocarbon spectra; however, quantitative mineral oil–in–water analysis requires certified reference materials and validated methods. Therefore, this product may be suitable for general or qualitative FTIR observations, but is not recommended for quantitative mineral oil–in–water analysis.

  2. What is the shelf life of this oil?

    1 answer
    1. This product is not assigned an expiration or retest date. Products that are robust in nature and extremely stable, in the original unopened container, are not assigned an expiration date or retest schedule. These products will have no dating reported on either the label or the Certificate of Analysis. Products that are not assigned an expiration or retest date are covered by a warranty period of 1 year from the date of product shipment.

  3. Can M8410 Sigma-Aldrich Mineral oil be used for the deparaffinization of FFPE samples, and is it compatible with downstream histological or molecular analyses like immunohistochemistry or RNA/DNA extraction?

    1 answer
    1. This product has not been validated for deparaffinization of formalin-fixed, paraffin-embedded samples. However, an article in "Applied Immunohistochemistry & Molecular Morphology" (2014 Sep;22(8):623-6) indicated that mineral oil appears to work well for deparaffinizing these samples and for DNA extraction.

  4. Hello,I´m looking for a mineral oil for sulfur pattern production. It´s difficult to find an oil in which the content of sulfur compounds appears in the COA. Can you inform if there´s a possibility of sulfur contamination in the production of mineral oil?

    1 answer
    1. The manufacturing process for this product is considered proprietary. Currently, there is no product option available that is tested for the presence of sulfur.

  5. ıs this suitable for preparation of carbon paste electrode?

    1 answer
    1. This material has not been validated in the process of making carbon paste electrodes. However, if the specifications (found below) meet your requirements, then there is no reason that they can't be used in this process.

  6. Hello, Does this product have an odour? I want to use it for an olfactory memory assay, so needs to be odourless

    1 answer
    1. According to Section 9 of the Safety Data Sheet, this is an odorless chemical.

  7. What is the composition of the oil? Where does it comes from?

    1 answer
    1. M8410 is a light oil. Mineral oil is a complex mixture of different hydrocarbons and its typical composition is as follows:

Paraffinic oils, based on n-alkanes, 15-60%
Naphthenic oils, based on cycloalkanes, 30%-60%
Aromatic oils, based on aromatic hydrocarbons, 3-30%
Light and heavy mineral oil differ in terms of composition, viscosity, and density.
Mineral oil with an alkyl chain carbon number of 15 to 25 is categorized as light oil whereas 25 to 50 as heavy oil.
Light oil has a density of 0.83–0.86 kg L-1 whereas heavy oil has 0.875–0.905 kg L-1
Light mineral oil is less occlusive than heavy mineral oil as it contains more cyclic (saturated) molecules
