Application

Collagen from chicken sternal cartilage has been used:

  • in enzyme–linked immunosorbent assay (ELISA).[1]
  • chondrocyte-mediated tissue production in vitro.[2]
  • induction, treatment, and assessment of collagenα induced arthritis (CIA). [3][4]
  • cell proliferation assay.[5]
  • as a coating for cell culture surfaces.[5]

Biochem/physiol Actions

Collagen breaks down metabolically in the body to release N-telopeptide, which is the N-terminus of collagen. There is also C-telopeptide, which is presumably the C-terminus. N-telopeptide is released in urine, and its detection in diagnostic tests is used to screen for osteoporosis.

Features and Benefits

This collagen has been tested in culture with mammalian cells to verify it is low in endotoxin content.

General description

Collagen is classified into a number of structurally and genetically distinct types. We use the nomenclature proposed by Bornstein and Traub. Do not confuse Sigma type designations with recognized collagen classification types.
Collagen is made of three polypeptide chains.[6] Type 2 collagen is ubiquitously present in the hyaline cartilage. It is a homotrimer made of three α1[7] chains.[6] Type 2 collagen is also predominant in nucleus pulposus of the vertebral disc and vitreous body of the eye.[7] The proteoglycan obtained from the chicken sternal cartilage is similar to type nine collagen structure.[8]

  1. Is this product sterilized? If not, how do we sterilize it? Thank you!

    1. This product is not sterile as received. Collagen solutions may be sterilized by dialysis in a 0.25% acetic acid and 0.5% chloroform solution. We do not recommend sterilizing the collagen solution by membrane filtration as we have found substantial protein loss by this method. Coating of tissue culture plastic dishes may be performed by air-drying. Dried coated dishes can be sterilized by exposure to UV light in a sterile culture hood or by rinsing with 70% ethanol.     1. The product C9301 gives 1 major band at approximately 150 KDa in the SDS-PAGE test.

  3. What is the recommended method for storing stock solutions of item C9301, Collagen from chicken sternal cartilage after reconstitution?

    1. Following reconstitution, store the solution in the refrigerator at 4°C. Stock solutions are stable for up to 6 months at 4°C. It is important to avoid freeze/thaw cycles of the solutions.
