General description

The ZytoLight® SPEC MET/CEN 7 Dual Color Probe (PL46, Z-2087-50/200) is a FISH probe designed for the qualitative detection of amplifications associated with the human MET gene and the alpha-satellite regions of chromosome 7 in formalin-fixed, paraffin-embedded (FFPE) specimens. This probe utilizes ZyGreen-labeled polynucleotides to detect MET gene amplifications at 7q31.2 (chr7:115,925,700-116,718,699, ~10.0 ng/µl) and ZyOrange-labeled polynucleotides for the chromosome 7 alpha satellites (D7Z1, 7p11.1-q11.1, ~1.5 ng/µl). It is intended for use with the ZytoLight FISH-Tissue Implementation Kit (Prod. No. Z-2028-5/-20).

Principle:

The ZytoLight® system uses fluorescence in situ hybridization (FISH) to detect and visualize specific nucleic acid sequences in formalin-fixed, paraffin-embedded (FFPE) specimens. Direct-labeled probes co-denature with complementary target DNA strands, facilitating hybridization. Unbound probes are washed away, and hybridized fragments are counterstained with DAPI.The ZytoLight® SPEC MET/CEN 7 Dual Color Probe includes:ZyGreen: Targets the MET gene region at 7q31.2 (excitation 503 nm/emission 528 nm).ZyOrange: Targets chromosome 7 alpha satellites (excitation 547 nm/emission 572 nm).FISH visualizes green signals for the MET gene and orange signals for chromosome 7 using a microscopy setup with exciter, dichroic, and barrier filters. This system is optimized for detecting genetic aberrations like amplifications and aneuploidies in tissue sections, eliminating the need for antibodies through direct labeling.

Probe Composition:

  • ZyGreen: Labeled polynucleotides (~10 ng/µl) targeting the MET gene region at 7q31.2 (chr7:115,925,700-116,718,699).
  • ZyOrange: Labeled polynucleotides (~1.5 ng/µl) targeting chromosome 7 alpha satellites (D7Z1).
  • Hybridization Buffer: Formamide-based hybridization buffer.

Application

For the qualitative detection of human MET gene amplifications and alpha satellites of chromosome 7 by fluorescence in situ hybridization (FISH)
The probe is designed for qualitative detection of sequences in the MET gene region and chromosome 7 centromere in formalin-fixed, paraffin-embedded (FFPE) tissue specimens by FISH. It is used in combination with the ZytoLight® FISH-Tissue Implementation Kit for specimen pretreatment, hybridization, and post-hybridization processing.

Analysis Note

In normal interphase nuclei, two green signals (for the MET gene) and two orange signals (for chromosome 7) are expected. In cases of amplification of the MET gene, multiple copies of the green signal or clusters will be observed.
