General description
The ZytoLight® SPEC MYC Dual Color Break Apart Probe (PL49, Z-2090-50/200) is a FISH probe designed for the qualitative detection of translocations involving the human MYC gene at 8q24.21 in cytologic and formalin-fixed, paraffin-embedded (FFPE) specimens, such as diffuse large B-cell lymphoma (DLBCL) and Burkitt lymphoma (BL). This probe utilizes a green probe targeting the distal region (chr8:130,373,051-130,930,673, ~10.0 ng/µl ZyGreen) and an orange probe for the proximal region (chr8:127,888,765-128,363,281, ~4.5 ng/µl ZyOrange). It is intended for use with the ZytoLight FISH Implementation Kits (Prod. No. Z-2028-5/-20 or Z-2099-20).
Principle:
The ZytoLight system uses fluorescence in situ hybridization (FISH) to detect and visualize specific nucleic acid sequences. Direct-labeled probes co-denature with their complementary target DNA strands, allowing hybridization. Unbound probes are washed away, and hybridized fragments are counterstained with DAPI, visualized using fluorescence microscopy with appropriate filters. The ZytoLight® SPEC MYC Dual Color Break Apart Probe includes: ZyGreen: Targets sequences distal to the MYC breakpoint at 8q24.21 (excitation 503 nm/emission 528 nm). ZyOrange: Targets sequences proximal to the MYC breakpoint at 8q24.21 (excitation 547 nm/emission 572 nm).FISH visualizes green and orange signals corresponding to the MYC gene regions, optimized for detecting 8q24.21 genetic aberrations in tissue sections and cytology specimens, enabling translocation detection without the need for antibodies.

Application
For the qualitative detection of translocations involving the human MYC gene at 8q24.21 by fluorescence in situ hybridization (FISH)
The probe is designed for qualitative detection of translocations involving the 8q24.21 region in cytologic or formalin-fixed, paraffin-embedded (FFPE) tissue specimens by FISH. It supports both cytology and tissue workflows using compatible ZytoLight® Implementation Kits.
Analysis Note
In normal interphase nuclei, two orange/green fusion signals are expected, indicating non-rearranged loci. In cases of translocation, a signal pattern with one orange signal and one separate green signal will be observed.
