General description
The ZytoLight® SPEC ERBB2/TOP2A/CEN 17 Triple Color Probe (PL52) is a direct-labeled FISH probe designed for the qualitative detection of amplifications involving the human ERBB2 and TOP2A genes, as well as the detection of chromosome 17 alpha satellites in formalin-fixed, paraffin-embedded (FFPE) specimens. It is intended to be used with the ZytoLight FISH-Tissue Implementation Kit.
Principle:
The fluorescence in situ hybridization (FISH) technique enables the detection and visualization of specific nucleic acid sequences. Fluorescently-labeled DNA fragments (FISH probes) co-denature and hybridize with complementary target DNA strands, followed by stringent washing to remove unbound fragments. The hybridized probes are visualized using a fluorescence microscope equipped with specific excitation and emission filters for the fluorochromes.In this assay, 10 µl of the probe is applied to pretreated formalin-fixed, paraffin-embedded (FFPE) sections. The sections are denatured for 10 minutes at 75 °C, hybridized overnight at 37 °C, and then washed and examined to detect signals: green for ERBB2, orange for TOP2A, and blue for CEN 17. The use of directly labeled DNA probes streamlines the process by eliminating the need for antibody detection.

- ZyGreen labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting the ERBB2 gene region on chromosome 17 (17q12-q21.1).
- ZyOrange labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting the TOP2A gene region on chromosome 17 (17q21.1-q21.2).
- ZyBlue labeled polynucleotides (excitation 418 nm/emission 467 nm) targeting the alpha satellite centromeric region D17Z1 of chromosome 17.
- Formamide-based hybridization buffer.
