General description

The ZytoLight® SPEC PTPRT/20q11 DualColor Probe is a probe that can detect deletionsof the long arm of chromosome 20.
The fluorescence in situ hybridization (FISH) technique allows for the detection and visualization of specific nucleic acid sequences in cell preparations. Fluorescently-labeled DNA fragments, so called FISH probes, and their complementary target DNA strands in the preparationsare co-denatured and subsequently allowed to anneal during hybridization. Afterwards, unspecific and unbound probe fragments are removed by stringency washing steps.
ZytoLight SPEC PTPRT/20q11 Dual Color Probe is composed of:

  • ZyGreen (excitation 503 nm/emission 528 nm) labeled polynucleotides (~10 ng/µl), which target sequences mapping in20q12* (chr20:40,807,040-41,419,634) harboring the PTPRT gene region
  • ZyOrange (excitation 547 nm/emission 572 nm) labeled polynucleotides (~4.5 ng/µl), which target sequences mapping in20q11.21* (chr20:30,301,019-30,719,110)
  • Formamide based hybridization buffer

Application

This probe is used for the qualitative detection of human PTPRT genedeletions and human 20q11 specific sequences byfluorescence in situ hybridization (FISH). It is designed to detect 20q deletions which causes various myeloiddisorders, e.g., myelodysplastic syndromes(MDS), acute myeloid leukemia (AML),and myeloproliferative neoplasms (MPNs).

Analysis Note

  • Hybridization signals of the probe appear green (PTPRT gene region) and orange (20q11.21 locus)
  • Normal situation: In interphases of normal cells or cells without a deletion involving the PTPRT gene region, two green and two orange signals appear
  • Aberrant situation: In a cell with deletion affecting the PTPRT gene region, a reduced number of green signals will be observed. Deletions affecting only parts of the PTPRT gene region might result in a normal signal pattern with green signals of reduced size
  • Overlapping signals may appear as yellow signals
  • Red blood cells present in the specimen might exhibit autofluorescence which hinders signal recognition.
