Beschreibung

General description

Ein zellpermeabler, potenter und selektiver Inhibitor der Akt1/Akt2-Aktivität (IC₅₀ = 58 nmol/l, 210 nmol/l und 2,12 µmol/l für Akt1, Akt2 bzw. Akt3, in In-vitro-Kinase-Assays). Die Hemmung ist Berichten zufolge von der Pleckstrin-Homologiedomäne (PH-Domäne) abhängig. Sie hat keine hemmende Wirkung gegen Akt ohne PH-Domäne oder andere eng verwandte Kinasen der AGC-Familie, PKA, PKC und SGK, selbst bei Konzentrationen von 50 µmol/l. Sie überwindet den durch Akt1/Akt2 vermittelten Widerstand gegenüber Chemotherapeutika in Tumorzellen und blockiert die basale und stimulierte Phosphorylierung/Aktivierung von Akt1/Akt2 sowohl in kultivierten Zellen in vitro als auch in Mäusen in vivo.
