Beschreibung

General description

Eine zellpermeable Purin-Verbindung, die als hochaffiner Aryl-Hydrocarbon-Rezeptor (AhR)-Antagonist dient (IC₅₀ = 62 nmol/l in Mäuseleberzytosol, das humanisierten AhR exprimiert) und dem selbst bei höheren Dosierungen jegliche Agonistenaktivität fehlt. Bindet Berichten zufolge mit hoher Affinität an die Ligandenbindungstasche von AhR und blockiert die Bindung einer Reihe von exogenen und endogenen Liganden. Interagiert nichtkovalent mit Ser³¹⁷, His²⁹¹ und Ser³⁶⁵ bei humanem und mit Ser³¹¹, His²⁸⁵ und Ser³⁵⁹ bei Maus-AhR. Unterdrückt die AhR-Transkriptionsaktivität durch Dioxin-Response-Element (DRE)-abhängige und unabhängig vermittelte Reaktionen in Zellen von Mensch und Maus (IC₅₀ = 8,5 nmol/l in HepG2-40/6-Zellen).
