Beschreibung

General description

Eine zellpermeable, tri-substituierte Pyrazolverbindung, die als starker und spezifischer Inhibitor der Integrin-verknüpften Kinase (ILK) (IC₅₀ = 600 nM) fungiert. Weist in-vitro nachweislich eine hohe vermehrungshemmende Potenz gegen Prostata- und Brustkrebszelllinien auf (IC₅₀ = 1 bis 2,5 µ;M), während es normale Epithelzellen verschont. Seiner Wirkung auf Krebszellen wird die Induktion der Autophagie und Apoptose zugeschrieben. Unterdrückt die ILK-vermittelte Phosphorylierung von Akt an der Stelle Ser473, wodurch die nachgelagerten Ziele, wie GSK-3b und die Myosin-Leichtkette, herunterreguliert werden. Verursacht zudem eine transkriptionelle Repression des Y-Box-Bindungsproteins 1 (YB-1) des Transkriptionsfaktors. Weist minimale Toxizität im Mausmodell auf.
