Beschreibung

General description

Eine zellpermeable Pyridinylfuranopyrimidin-Verbindung, die als potenter und mit ATP konkurrierender Inhibitor von DNA-PK, PI3-K und mTOR (IC₅₀ = 2, 8, 88, 48, 150, 26, 20 bzw. 83 nM für DNA-PK, p110α;, p110β;, p110δ;, p110γ;, PI3-KC2β;, mTORC1 bzw. mTORC2) wirkt. Sie hemmt ATR und ATM nur bei wesentlich höheren Konzentrationen (IC₅₀ = 850 bzw. 920 nM) und legt selbst bei Konzentrationen von bis zu 10 µ;M geringe Aktivität gegenüber einem Panel von über 40 anderen Kinasen an den Tag. Blockiert nachweislich wirksam den PI3-K/Akt-Signalweg und die Zellproliferation in Gliomzelllinien sowohl in vitro als auch in vivo. Eine 10-mM-Lösung (2 mg/574 µ;l) von PI-103 (Katalog-Nr. <;A href="https://./?[islink]ViewProductDetail-BySKU&;#124;&;#124;SKU&;#124;EMD_BIO-528101&;#124;[/islink]">;528101<;/A>;) in DMSO ist ebenfalls erhältlich.
