General description

Benzonase® Nuklease ist eine gentechnisch veränderte Endonuklease aus Serratia marcescens. Sie baut alle DNA- und RNA-Formen (einzelsträngig, doppelsträngig, linear und zirkulär) ohne proteolytische Aktivität ab. Sie ist über ein breites Spektrum von Bedingungen wirksam und weist eine außerordentlich hohe spezifische Aktivität auf. Das Enzym dient dem kompletten Verdau von Nukleinsäuren in 5′-Monophosphat-terminierte Oligonukleotide mit einer Länge von 2 bis 5 Basen (unterhalb der Hybridisierungsgrenze). Dies ist ideal für die Entfernung von Nukleinsäuren aus rekombinanten Proteinen und ermöglicht die Einhaltung der FDA-Richtlinien für Nukleinsäurekontamination. Aufgrund ihrer Fähigkeit, Nukleinsäuren rasch zu hydrolysieren, ist Benzonase eine ausgezeichnete Wahl zur Reduzierung von Viskosität, Verkürzung der Verarbeitungsdauer und Erhöhung der Proteinausbeute. Das Enzym ist beispielsweise mit BugBuster und PopCulture Proteinextraktionsreagenzien kompatibel und kann daher zusammen mit diesen Reagenzien zugegeben werden, um bei E. coli-Extrakten Viskosität zu eliminieren und Nukleinsäuren zu entfernen.

Das Enzym besteht aus zwei Untereinheiten von je 30 kDa. Es ist zwischen pH-Wert 6 und 10 und von 0–42 °C funktionsfähig und erfordert 1–2 mmol/l Mg2+ zur Aktivierung. Das Enzym ist auch in Gegenwart von ionischen und nichtionischen Detergenzien, Reduktionsmitteln, PMSF (1 mmol/l), EDTA (1 mmol/l) und Harnstoff aktiv (die relative Aktivität hängt von den spezifischen Bedingungen ab). Die Aktivität wird durch >150 mmol/l monovalente Kationen, >100 mmol/l Phosphat, >100 mmol/l Ammoniumsulfat oder >100 mmol/l Guanidin-HCl gehemmt.

Benzonase-Nuklease ist in den Qualitätsstufen reinst (>99 % mittels SDS-PAGE) und rein (>90 %) in einer Standardkonzentration von 25–29 E/µl und in einer hohen Konzentration (HC) von 250 E/µl erhältlich. Beide Zubereitungen sind frei von nachweisbaren Proteasen und weisen eine spezifische Aktivität von > 1 × 106 E/mg Protein auf. Der >99%ige Reinheitsgrad wird auf Endotoxine getestet und enthält <0.25 EU/1000 units. The product is suppliedin 50% glycerol.Store at -20°C.

  1. How long can the product be stored at -20°C?

    1. This product is not assigned a firm expiration date. Each lot is assigned a recommended retest date of approximately 1 year from the date of quality release. If sufficient stock remains and the material continues to meet product specifications upon reassay, the retest period is typically extended. While considered stable, the material may be subject to degradation over time. The lot-specific retest date is reported on the Certificate of Analysis.

  2. Can proteinase K be utilized to inactivate Benzonase? This is to prevent further DNA degradation post-sampling. Would it be adequate to treat it with a final concentration of 10 mg/ml of proteinase K for 20 minutes at 55°C?

    1. There is currently no available data on the conditions required for inactivating benzonase by proteinase K. However, it is known that benzonase can be reversibly inactivated by metal ion chelators, such as EDTA and EGTA. Irreversible inactivation can also be achieved through extreme conditions (100 mM NaOH at 70°C for 30 minutes).

    1. Benzonase can be inactivated through various methods such as treatment with NaOH, EDTA and warming with EDTA at 70°C, or guanidine HCl. However, it is important to note that a guanidine HCl concentration exceeding 100 mM can completely inhibit the enzyme activity. Additionally, while an EDTA concentration of 1mM partially inhibits Benzonase endonuclease, a concentration of 5 mM EDTA leads to a loss of over 90% of enzyme activity due to the complexing of the essential Mg2+ ions.
