Application
Accumax hat sich bei der Dissoziation verklumpter Zellen in Suspensionskulturen aus Hybridomen, CHO-, BHK-, 293-, COS- und Sf9-Zellen als effektiv erwiesen. Die Lösung hat bei der dissoziierenden Zellzählung sowie bei viralen Transfektions-Assays, Zellsortierung, Durchflusssymmetrie und Bioreaktor-Scaleup außerordentlich gute Leistungen gezeigt. Sie wurde auch schon zur Herstellung von Einzelzellsuspensionen aus Kulturen mit verklumpten Zellen für die korrekte Zellzählung, Ablösung von Zellen aus Primärgewebe und routinemäßige Zellablösung von Standard-Kunststoffmaterialien und haftbeschichteten Kunststoffmaterialien in Gewebekulturen verwendet.
Anwendungsbereiche der TM=[„Accumax“]-Lösung:
- Ablösung von einwändigen Kohlenstoff-Nanoröhrchen von der extrazellulären Matrix (ECM) in Epithelzellen
- Dissoziation von pigmentierten Netzhaut-Epithelzellen vor dem Passagieren
- Dissoziation von Langerhans-Inseln
Biochem/physiol Actions
Accumax ist eine Zellablösungslösung, die aus proteolytischen und kollagenolytischen Enzymen besteht. Jede Flasche enthält 100 ml 1x Accumax-Enzyme in Dulbecco′s phosphatgepufferter Kochsalzlösung (DPBS). In Accumax sind keine Säuger- oder Bakterienprodukte zu finden.

Preparation Note
Dieses Produkt wird als gebrauchsfertige und sterile gefrorene Flüssigkeit geliefert. Accumax sollte vor Gebrauch bei 37 °C aufgetaut und einer Probe aus verklumpten Zellen im selben Mengenverhältnis hinzugefügt werden. 5 - 10 Minuten bei 37 °C inkubieren und Zellen ganz normal zählen.
Zubereitet in Dulbeccos PBS (0,2 g/l KCl, 0,2 g/l KH2PO4, 8 g/l NaCl und 1,15 g/l Na2HPO4). Für Informationen über die Produktanwendung siehe das Produktdatenblatt auf der Website.

This Accumax™ solution does not contain mammalian- or bacterial-derived products. It is not a GMP grade. This product is assigned a quality level of 200. Products with quality level of 200 are not suitable for GMP. Products with quality levels 400 or higher are suitable for GMP.

What is the appropriate cell dissociating product to be used for ES cells that will be injected into blastocysts?

Accutase®, product A6964, or Accumax™ (Accutase® with DNase), product A7089, will work well for dissociation of ES cells. The Accutase® product has protease and dispase activity to dissociate the cells. The Accumax™ product also has DNase to limit the aggregation that is caused by DNA released from ruptured cells during the dissociation/scraping.Please see the reference and link below for more information on the use of Accutase® with ES cells:Mol Reprod Dev. 2008 May;75(5):818-27
