Application
Chondroitinase ABC von Sigma wird zum Aufschluss von O-gebundenen Chondroitinsulfat-ähnlichem Glykosaminoglykan (CSGAG) eingesetzt. Das Enzym wird auch zum Aufschluss von Glykosaminoglykanketten in Proteoglykanen während der Bildung und Expression eines rekombinanten Versican-Gens in Säugetiersystemen eingesetzt. Darüber hinaus wird es zum Aufschluss von Chondroitinsulfat in den Proteoglykankomplexen eingesetzt. Das Enzym wird auch zum Entfernen von Glykanketten aus Aggrecan verwendet, wodurch die axonwachstumshemmende Wirkung von Aggrecan entfernt wird.
Biochem/physiol Actions
Chondroitinase ABC katalysiert den eliminativen Abbau von Polysacchariden, die (1-4)-β-D-Hexosaminyl- und (1-3)-β-D-Glucuronosyl- (oder (1-3)-α-L-Iduronosyl)-Bindungen an Disaccharide enthalten, die 4-Desoxy-β-D-gluc-4-enuronosyl-Gruppen enthalten. Es wirkt auf Chondroitin-4-sulfat, Chondroitin-6-sulfat und Dermatansulfat. Es wirkt langsam auf Hyaluronat. Die Molekülmasse beträgt etwa 120 kDa, wobei zwei nicht identische Untereinheiten Molekülmassen von 86 kDa bzw. 32 kDa aufweisen. Der optimale pH-Wert beträgt 8,0 für Chondroitinsulfat und 6,8 für Hyaluronsäure. Die optimale Temperatur beträgt 37 °C. 1 mM Zn2+ dient als Inhibitor, während 0,05 M Acetat ein Aktivator des Enzyms sind.

Preparation Note
In einer 0,01 % wässrigen Rinderserumalbuminlösung (BSA-Lösung) rekonstituieren. Nachfolgende Verdünnungen können zu einem Puffer mit 50 mM Tris, pH-Wert 8,0, mit 60 mM Natriumacetat und 0,02 % BSA gemacht werden. Die Lösungen sollten frisch zubereitet werden.

Eine Einheit setzt bei einem pH-Wert von 8,0 und 37 °C je Minute 1,0 μmol einer Mischung aus 2-Acetamid-2-desoxy-3-O-(β-D-gluc-4-en-pyranosyluronsäure)-4-O-sulfo-D-galactose und 1,0 μmol 2-Acetamid-2-desoxy-3-O-(β-D-gluc-4-en-pyranosyluronsäure)-6-O-sulfo-D-galactose aus Chondroitinsulfat aus Haifischknorpel frei.
