Citratpuffer, pH 6,0, 10×, Antigen-Rückgewinnung wird verwendet für die wärmeinduzierte Antigen-Rückgewinnung aus formalinfixierten, in Paraffin eingebetteten (FFPE) Gewebeschnitten vor der Anwendung von Antikörpern oder DNA-Fluoreszenz-in-situ-Hybridisierung. In den Immunhistochemie (IHC) maskieren die gebräuchlichsten Fixative, zum Beispiel Formalin, aufgrund ihrer Vernetztheit Antigene (zelluläre, membranbindende und nukleäre). Daraus ergibt sich in der IHC eine schlechte oder ganz fehlende Färbung. Die Anwendung von Citratpuffer, pH 6,0, oder anderer Lösungen für die Antigen-Rückgewinnung auf FFPE-Gewebeschnitte verbessert den Zugang von Antikörpern zu den Antigenen des Gewebes.
Preparation Note
Das Produkt bei 2–8 °C lagern. Zur Vermeidung einer möglichen Fällung nicht gefrieren.
Den Citratpuffer, pH 6,0, 10x, Antigen-Rückgewinnung 10-fach mit Wasser verdünnen, um eine 1x Arbeitslösung herzustellen, z. B. 10 ml 10x-Konzentrat mit 90 ml Wasser verdünnen.
Other Notes
Gelegentlich hat der Puffer einen leicht gelblichen Farbstich. Dies ist auf das Konservierungsmittel zurückzuführen und beeinträchtigt nicht die Funktion des Puffers.
Den vorbereiteten 1′-Citratpuffer, pH 6,0, Arbeitslösung bei 2–8 °C lagern.

Is this a mixture from trisodium citrate and citric acid? I have looked through the SDS and the mixture of the chemical is <0.025% of 5-Chloro-2-methyl-4-isothiazolin-3-one and another chemical. I can't seem to find how they would make up a citrate buffer

The specific formulation is considered proprietary, however, the final citrate concentration is 0.1M. The SDS lists only the potentially hazardous ingredients. The 5-chloro-2-methyl-4-isothiazolin-3-one and 2-methyl-2H-isothiazol-3-one (3:1) listed is essentially ProClin which acts as a preservative. What molarity does this buffer have?

Product C9999 is provided at a concentration of 0.1 M. It'll be 10 mM when diluted. The solution contains 116.25 mM Tri-NaCitrate, 10.25 mM EDTA, 0.5 % ProClin, and other proprietary materials.

Is this Citrate buffer already included Tween 20?

The composition of this buffer is considered proprietary. However, citrate solutions commonly used for antigen retrieval do not contain detergents. This product does contain a preservative which may contribute to the color of the solution. This does not effect the performance of the product in any way. Please see the link below to reivew a general antigen retrival protocol using Citrate Buffer. Note that Tween 20 is used in the wash solution following treatment with Citrate Buffer.
