Application

Digoxin-BSA bovine is a cardioactive glycoside which can increase intracellular calcium concentration through Na+/K+-ATPase pump inhibition. Digoxin has been used in treatments of heart related disease and also studies of cancer treatments. It has shown the ability to inhibit prostate cancer cell growth, but has also shown a reduction in the efficiency of chemotherapy (reduced drug absorption) by increasing the amount of P-glycoprotein (Pgp).

Questions

1–2 of 2 Questions
  1. What solvent should be used for this product reconstitution? Any specific conditions?

    1 answer
    1. Quality testing of the product does not include the determination of its solubility. Literature indicates the product can be dissolved in DMSO and stored at 2-8°C for 2 weeks or 6 months at -80°C. Because solubility testing is not performed, this claim has not been validated.

      Helpful?

  2. Is Digoxin covalently conjugated to BSA? Is there any reference available to confirm this information?

    1 answer
    1. The preparation involves the use of the digoxin-hemisuccinate-BSA conjugate, which means that the digoxin is modified to the hemisuccinate and then linked to the BSA, likely through its epsilon-amino group. Further information can be found in the literature: Biol. Pharm. Bull., 23, 906 (2000).
