General description

Deoxyribonucleic acid, single stranded from salmon testes is a ready-to-use solution of high quality, sonicated, single-stranded template DNA isolated from the testes of salmon.

Application

Deoxyribonucleic acid, single stranded from salmon testes has been used for/as:

• in situ hybridization histochemistry of rat brain, human kidney sections and renal biopsy tissues
• preparing standard curve of DNA
• prehybridization solution in southern and northern hybridizations
• dilution of the cDNA
• fluorescent in-situ hybridization (FISH)
• ChIP analysis
• a standard to analyse the degraded nucleosides of genomic DNA by high performance liquid chromatography
• a component of hybridization solution in FISH
• a control in DNA-DNA hybridization experiment between various bacterial species

Als Blockierungsmittel bei Northern und Southern Blotting.

Bei Hybridisierungen von Nukleinsäuren tragen viele Faktoren zum Signal-Rauschen-Verhältnis bei. Dazu gehören unter anderem die Gegenwart eines Lösungsmittels (Formamid), Temperatur und Länge der Hybridisierung, Menge der Hybridisierungslösung, Grad und Methode der Agitation, Verwendung von Blockierungsreagenzien, Konzentration und spezifische Aktivität der Sonde, Verwendung von molekularen Wirkstoffen zur Steigerung der Reassoziationsrate der Nukleinsäuren sowie der Grad an Strenge, die beim Waschen der Membran herrschte.

Um jede nicht-spezifische Hybridisierung der Sonde an ein Substrat zu verringern, müssen Blockierungsmittel eingesetzt werden. Im Allgemeinen wird dies mit einer Kombination aus Blockierungsreagens, Reinigungsmittel und denaturierter, fragmentierter DNA erreicht. Sigma bietet geschallte, denaturierte DNA von vielen verschiedenen Spezies, die bei Northern und Southern Blotting und bei anderen Methoden für die Hybridisierung von Nukleinsäuren als Blockierungsmittel verwendet werden können.

Suitable for use as a blocking agent in Southern hybridizations.

Preparation Note

This DNA is phenol-chloroform extracted, ethanol precipitated, and sonicated to produce single-stranded fragments which comigrate with the 587 and 831 base pair marker fragments.

Other Notes

DNA in solution will reanneal on standing at room temperature so it is recommended to boil the solution for 10 minutes and then cool on ice for at least 5 minutes prior to use.
