Jeder Milliliter komplettes Freund-Adjuvans enthält 1 mg Mycobacterium tuberculosis (H37Ra, ATCC 25177), durch Hitze abgetötet und getrocknet, 0,85 ml Paraffinöl und 0,15 ml Mannidmonooleat.
Application
Antigene (vorzugsweise in Kochsalzlösung) werden normalerweise zwecks Bildung einer Emulsion mit einem Adjuvans gleichen Volumens gemischt.
Das Freund-Adjuvans kann zur Herstellung von Wasser-in-Öl-Emulsionen von Immunogenen verwendet werden. Antigene in Wasser-in-Öl-Emulsionen stimulieren starke und langanhaltende Antikörperreaktionen, die auf die langsame Freisetzung von Antigen zurückgeführt werden können.
Biochem/physiol Actions
Die Myobakterien im kompletten Freund-Adjuvans locken Makrophagen und andere Zellen zur Injektionsstelle, wodurch die Immunreaktion gesteigert wird. Aus diesem Grund wird das komplette Freund-Adjuvans für initiale Injektionen verwendet und das inkomplette Freund-Adjuvans für nachfolgende Wiederholungsinjektionen.
