General description
Hämoglobin ist die Hauptkomponente von Erythrozyten und für deren rote Farbe verantwortlich. Seine normale Konzentration in den Erythrozyten ist 34 %. Hämoglobin ist ein globuläres Protein mit α- und β-Ketten, die 141 bzw. 146 Aminosäuren enthalten. Es existiert als Tetramer, wobei jeder Monomer einen heterozyklischen Porphyrinring besitzt, in dem Eisen das zentrale Ion ist.
Application
Die Löslichkeit von α-Elastin wird bei der Entwicklung von Elastin-mimetischen Biomaterialien genutzt.
Hämoglobin aus Rinderblut hat folgende Anwendungen:
- Erstellung von Standardkurven für die Quantifizierung intraparenchymaler und parenchymaler Blutungen in Rückenmarkshomogenat unter Verwendung von Drabkin-Assay, Quadrupol-Ionenmobilitäts-Flugzeit-Massenspektrometrie
- Herstellung von molekular aufgedruckten Polymeren (MIP) zur Nachahmung von Polyethylenglycol (PEG) mit hohem Molekulargewicht in Kristallisierungsstudien

Sauerstofftransporter. Das Gleichgewicht zwischen Fe2+ und Fe3+ ist ein physiologischer Indikator für die Oxygenierung des Blutes. Desoxygeniertes Hämoglobin liefert einen Rückführungsmechanismus, indem Nitrit zu NO reduziert wird, d. h. zu einem Vasodilatator, der den Blutfluss an sauerstoffarmes Gewebe erhöht.
Hämoglobin ist aufgrund seiner Fähigkeit zum Transport des Sauerstoffes von der Lunge zu den Organgeweben und Austausch des Kohlenstoffdioxids, das wichtigste respiratorische Protein von Wirbeltieren. Rinderhämoglobin wird verwendet, um Hämoglobin-Vesikel (HbV) für den Sauerstofftransport zu erzeugen. Es ist thermisch stabil und zeigt im Vergleich zu menschlichem Hämoglobin eine hohe Affinität zu Sauerstoff. Rinderhämoglobin interagiert mit synthetischen und Azofarbstoffen. Polymerisierte Formen von Rinderhämoglobin werden zur Behandlung von autoimmunhämolytischer Anämie empfohlen.

I want to create hemoglobin standards to use in a colorimetric assay. Can I create hemoglobin concentrations by weighing specific amounts of lyophilized hemoglobin powder and reconstituting with TS saline? Then process in assay after adding reagent?

What is the difference between H2500 and H2625 when making Chocolate agar with bovine hemoglobin? Although they share the same CAS and Specification Sheet, there is a significant difference in price between the two.

Products H2500 and H2625 are not suitability tested for use in producing Chocolate Agar. The key differences between H2500 and H2625 lie in their purity and intended uses. H2500 is a purer product, suitable for MALDI-TOF, and associated with a companion product for HPLC analysis of hemoglobin variants. It undergoes two steps of ammonium sulfate precipitation, filtration, dialysis, and lyophilization. On the other hand, H2625 is not suitability tested for use as a protease substrate and lacks the ammonium sulfate precipitation step. These distinctions likely contribute to the significant price difference between the two products.

The website indicates that the product H2500 is mainly methemoglobin. A search of the website did not uncover any products marketed as either ferrohemoglobin or oxyhemoglobin.
