General description

Poly-L-lysine is a positively charged amino acid polymer with approximately one HBr per lysine residue. The hydrobromide allows the poly-L-lysine to be in a crystalline form soluble in water. A small amount of product may be found in the beta structure because the HBr interferes with hydrogen bonding between amino and either the carboxyl groups or N or O containing moieties. Poly-L-lysine hydrobromide is a nonspecific attachment factor in promoting cell adhesion to the substratum.

Application

Recommended as a cell culture substratum when using 0.5 ml of a 0.1 mg/ml solution to coat 25 cm². Optimal conditions for attachment must be determined for each cell line and application.
Poly-L-lysine hydrobromide has been used to adhere the dissociated cells of cortical tissues into the culture plates. It has also been used to treat slides, to fix samples on it for immunohistochemical analysis.

Biochem/physiol Actions

Poly-L-lysine is employed as a coating substance to facilitate improved cell attachment to culture surfaces. Research indicates that plates coated with poly-L-lysine (PLL) exhibit an accelerated cell growth rate. When mesenchymal stem cells (MSCs) are cultured on PLL-coated substrates, it helps preserve their stemness properties and prevents senescence. PLL-coated plates promote enhanced MSC growth and up-regulation of genes associated with cell adhesion, differentiation, proliferation, and signaling. Furthermore, porous PLL-coated surfaces have been shown to enhance cell proliferation and support the osteogenic differentiation of MSCs.

Poly-L-Lysin ist ein positiv geladenes Aminosäurepolymer mit etwa einem HBr pro Lysin-Rest. Das Hydrobromid ermöglicht die kristalline, wasserlösliche Form des Poly-L-Lysins. Eine geringe Produktmenge kann in der β-Struktur zu finden sein, da das HBr die Wasserstoffbindung zwischen Aminogruppen und den Carboxylgruppen oder N oder O enthaltenden Gruppen beeinträchtigt.
