General description
A-83-01 kann die Kontraktion von Verbrennungswunden wirksam verhindern, ohne den Wundverschluss zu behindern, da es den durch den transformierenden Wachstumsfaktor β (TGF-β) induzierten Anstieg der Myofibroblastenpopulation hemmen kann. Darüber hinaus kann er TGF-β1-abhängige Krebsmetastasen durch Unterdrückung des epithelialen mesenchymalen Übergangs (EMT) bei Tieren verhindern.
Application
A 83-01 wurde verwendet:
- im Kulturmedium für die Organoidbildung aus dissoziierten Tumorzellen
- als Komponente in Dulbecco′s Modified Eagle Medium/Nutrient Mixture F-12 (DMEM/F12) zur Kultivierung humaner epidermaler Stammzellen (EpSCs)
A 83-01 wird als Inhibitor des TGF (Transforming Growth Factor) und des Kinase-Typ-1-Rezeptors verwendet.
Biochem/physiol Actions
A 83-01 ist ein Inhibitor von TGFβ-Kinase bzw. ALK 5 (Activin Receptor-like Kinase) (IC50 = 12 nM), der die Phosphorylierung von Smad2/3 verhindert und das von TGFβ induzierte Wachstum hemmt. A 83-01 blockiert die Phosphorylierung von Smad2 und hemmt die von TGFβ induzierte epithelial-mesenchymale Transition. Außerdem hemmt A 83-01 die durch den TGFβ-Typ-I-Rezeptor ALK-5, den Activin-Typ-IB-Rezeptor ALK-4 und den nodalen Typ-I-Rezeptor ALK-7 induzierte Transkriptionsaktivität. A 83-01 induziert die Expansion von Nkx2.5-eGFP (+) Zellen bei Neugeborenen.
