General description
Trehalose ist ein überall in der Natur anzutreffendes nicht reduzierendes Disaccharid. Es wird als Konservierungsmittel für Lebensmittel und zum Erhalt von Enzymen, Impfstoffen, Zellen und vielen anderen Molekülen verwendet. In wirbellosen Tieren ist Trehalose als Hämolymphzucker vorhanden.
Application
D-(+)-Trehalosedihydrat wird beim Einfrieren von Kaninchen-Oozyten (langsame Einfriermethode) verwendet. Darüber hinaus wird es bei der Solid Surface Vitrification (SSV) von in vitro gereiften Oozyten eingesetzt. Ist für den Einsatz in einer Vielzahl von Zellgefriermedien als Kälteschutzmittel geeignet.
Biochem/physiol Actions
Trehalose ist für die Embryogenese, den Kohlenstoff-Stoffwechsel, die Photosynthese und die Pflanzenblüte von wesentlicher Bedeutung. Es existiert vornehmlich unter den Spezies, die die Anhydrobiose (ein Leben ohne Wasser) unterstützen. Es dient als Kohlenhydratreserve in Mikroorganismen und schützt diese vor nachteiligen Bedingungen wie Wärme, Kälte, Dehydration, Austrocknung und Oxidation. Trehalose wirkt als chemisches Chaperon mit Auswirkungen auf die Proteinfaltung. Sie verhindert bekanntermaßen die Amyloiderzeugung und die Anhäufung von β-Amyloid in vitro. Ein ähnlicher Effekt der Trehalose wurde mit der Huntingtin-Proteinbindung beobachtet. Folglich könnte sie als Schutzmittel bei der Alzheimer- und Huntington-Krankheit eingesetzt werden.

We would like to dissolve trehalose in water in bulk and store it. Does dissolved trehalose loose its stability after an extended storage time? Which storage temperature would you recommend?

The solution stability and storage of this product has not been tested, however other commercial sources offer 1M Trehalose solutions that are stored at ambient temperature or refrigerated (filter-sterilized). Trehalose is added to several of less stable protein preparations as a carrier and/or cryoprotectant. An example would be EGFR, E2645. This lyophilized product contains 10% Trehalose. Upon reconstitution the product can be stored for up to 1 year at -70 deg C. Given the information above, Trehalose may be quite stable in solution.

Yes, it should work as a substitute for protein in a freezing media. It is commonly used at a final concentration of 5% (50 mg/mL). Alternatively, one could use methylcellulose.
