KH domain-containing RNA-binding protein QKI (UniProt: Q96PU8; also known as Protein quaking, Hqk, HqkI) is encoded by the QKI (also known as HKQ) gene (Gene ID: 9444) in human. The gene QKI (quaking) is mapped to human chromosome 6q26. It is a RNA reader protein, which recognizes and binds specific RNAs, and belongs to signal transduction and activation of RNA (STAR) protein family. QKI acts by regulating pre-mRNA splicing, mRNA export, mRNA stability and protein translation, and is itself, regulated by alternative splicing. QKI contains a KH RNA-binding domain flanked by two QUA domains, which facilitate dimerization and RNA interaction, crucial for its regulatory functions in alternative splicing, mRNA stability, and translation. Six isoforms of QKI have been described that are produced by alternative splicing but, exists predominantly as three major isoforms; QKI-5, QKI-6, and QKI-7, each differing in their C-terminal regions and subcellular localization, primarily within the nucleus and cytoplasm. QKI is expressed in the frontal cortex of brain, but is shown to be down-regulated in the brain of schizophrenic patients. Aberrant expression of QKI has been implicated in various diseases, including multiple sclerosis, schizophrenia, and certain cancers, where it may act as either a tumor suppressor or an oncogene depending on the context. Mouse homolog of QKI participates in neural development and myelination. Deficiencies in QKI are linked to severe demyelination. It also controls the production of circRNAs during epithelial-mesenchymal transition. In lung cancer cells, it suppresses proliferation by regulating alternative splicing of NUMB (protein S171) gene. QKI has also been demonstrated as a transcriptional co-activator beyond its canonical function as a RNA-binding protein. Clone 1J10 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects QKI. It targets an epitope within 19 amino acids from the internal region. This antibody is predicted to recognize QKI-6, QKI-7, and QKI-7B isoforms.

Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Angaben zur Herstellung

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
