Description générale

23.15 kDa (Proform) and 20.74 (mature form) calculated. ~27 kDa reported in UCHT1 immunoprecipitate (Poggi, A., et al. (1996). Int. Immunol. 8(12):1947-1953).
T-cell surface glycoprotein CD3 epsilon chain (UniProt P07766; also known as CD3-epsilon, CD3e, T-cell surface antigen T3/Leu-4 epsilon chain) is encoded by the CD3E (also known as T3E) gene (Gene ID 916) in human. The T cell receptor or TCR is a plasma membrane protein complex responsible for recognizing antigens bound to major histocompatibility complex (MHC) on antigen presenting cells. In 95% of T cells, the TCR is composed of a disulfide-linked alpha (α) and beta (β) dimer, while among 5% of T cells the TCR is composed of a gamma (γ) and a delta (δ) dimer. T cells are activated when their surface TCRs engage with antigen/MHC complex on antigen presenting cells. Each TCR alpha and beta chain contains a variable (V) and a constant (C) extracellular immunoglobulin-like domain, followed by a transmembrane region and a short cytoplasmic tail. The alpha and beta chains have very short cytoplasmic tail and rely on the CD3 (cluster of differentiation 3) co-receptor complex for transducing signaling further intracellularly. The CD3 co-receptor complex is composed of a CD3γ chain, a CD3δ chain, and two CD3ε chains. The CD3 complex, TCR dimer, and a ζ-chain together comprise the functional TCR complex. The CD3 epsilon chain is initially produced with a signal peptide (a.a. 1-22), the removal of which yields the mature chain (a.a. 23-207) with an Ig-like (a.a. 32-112) domain-containing extracellular region (a.a. 23-126), a transmembrane segment (a.a. 127-152), and a cytoplasmic tail (a.a. 153-207), which contains the immunoreceptor tyrosine-based activation motif or ITAM (a.a. 178-205) that is essential for mediating ZAP-70 interaction and transducing TCR signaling.

Immunocytochemistry Analysis: A representative lot was employed to determined the plasma membrane and cytoplasmic CD3 immunoreactivity in T cell populations and T-ALL blast cells by flourescent immunocytochemistry (Campana, D., et al. (1987). J. Immunol. 138(2):648-655).

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Evaluated by Flow Cytometry in human PBMC.

Flow Cytometry Analysis: 0.1 µg of this antibody detected CD3e in 1X10E6 human PBMC.
