Application

Le tampon RIPA a été utilisé comme réactif de lyse cellulaire dans les études expérimentales suivantes :
  • Phénotypage profond du tissu périphérique au moyen des fibroblastes, facilitant la stratification des troubles mécaniques dans la maladie de Parkinson sporadique[1]
  • Démonstration de l'inhibition de la phosphorylation oxydative (OXPHOS) qui peut activer la nécroptose dépendante de la protéine MLKL (Mixed Lineage Kinase domain-Like protein) dans les cellules épithéliales pulmonaires humaines[2]

Le tampon RIPA permet de lyser les cellules et de solubiliser les protéines de manière efficace, tout en évitant la dégradation des protéines et les interférences avec l′activité biologique et l′immunoréactivité des protéines. Le tampon RIPA donne aussi un faible bruit de fond dans les analyses d′immunoprécipitation et les analyses moléculaires du type « pull-down ». Compatible avec les gels d′affinité EZview.

Réduit au minimum les interactions par liaison protéique non spécifique pour garantir un faible bruit de fond.
Un millilitre du tampon RIPA permet de lyser les cellules présentes sur une boîte de Pétri de 100 mm (0,5 à 5 × 107 cellules) pour les lignées de cellules de mammifères les plus adhérentes.

General description

Le tampon pour analyse par radio-immunoprécipitation (tampon RIPA) est un tampon de lyse permettant de lyser rapidement et efficacement les cellules et de solubiliser les protéines issues des cultures de cellules mammifères adhérentes et en suspension.
Le tampon RIPA (Radio-Immunoprecipitation Assay) se présente sous la forme d'une solution prête à l'emploi ne nécessitant aucune préparation. Il minimise les interactions d'adsorption protéique non spécifiques afin de limiter le bruit de fond sans pour autant empêcher la plupart des interactions spécifiques, permettant ainsi l'étude des interactions protéine/protéine d'intérêt. Ce tampon est très utilisé dans des applications comme l'immunoprécipitation, car ses constituants n'affectent pas la plupart des anticorps et des antigènes protéiques.

  1. we bought a bottle of RIPA buffer and want to make alliquots of this bottle. Can we store the alliquots of the buffer by -20? Or is it better to keep them between 2°C-8°C ?

    1. It is recommended to store the product at 2–8C. Studies have not been performed on aliquots and to determine if refrigerated storage will extend the shelf life of this product. We would recommend searching the general research literature or contacting researchers who routinely use this product to see if solution stability information is available. If a recommended storage is not given in the literature, it is usually best to make solutions fresh for use. If you decide to store the aliquots at -20°C, we would recommend validating activity of the supplemented buffer after freezing and avoid any thawing/freezing cycle.     1. Regarding stability in the recommended temperature of 2-8 °C, this product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one year warranty from the date of shipment.     1. R0278 can be used on tissue, if it is well homogenized.

  8. How does Product R0278, RIPA buffer, compare to Product No. C2978 (CelLytic M)?

    1. RIPA buffer contains SDS, which is considered to be a harsh detergent; it can denature many proteins. C2978 is a buffered solution (pH 7.6) that contains a mild detergent; it is non-denaturing for most proteins.  The exact formulation of C2978, however, is proprietary.

  9. I left the bottle of Product R0278, RIPA buffer, on my bench for about a week. Can I still use it?

    1. Yes, you should be able to use this product.  Although we recommend storage in the refrigerator at 2-8°C for long-term storage, Product No. R0278 may be kept at ambient temperature (our normal shipping condition) for a short time (approximately 2 weeks).

  10. Does Product R0278, RIPA buffer, contain any protease inhibitors or phosphatase inhibitors?

    1. This buffer does not contain any protease or phosphatase inhibitors, however, they may be added to insure against possible proteolysis.
