General description

A cell-permeable, photo-stable nitric oxide (NO) fluorescent indicator with a detection limit of ~10 nM. Intracellularly releases DAR-4M (Cat. No. 251760) by the action of esterases. Reacts with NO, in the presence of O2, resulting in a triazolo-rhodamine analog (DAR-4M T) that exhibits about 840-fold greater fluorescence quantum efficiency. The fluorescence intensity is not dependent on pH.

Questions

  1. The product is provided in solution form but no information is provided about its volume. Please provide the volume and molarity of the solution. Also what would be the recommended way of dilution - in DMSO or directly in HBSS/PBS/media?

    1 answer
    1. This product is prepared in DMSO at a concentration of 3.125 mg/mL, which equates to approximately 5 mM. The volume of the 1 mg package is 0.320 mL. Prior to use, dilute the product 500-fold in 0.1 M phosphate buffer, pH 7.4. Other aqueous buffers such as HBSS would also be suitable.
Note that the presence of BSA and phenol red may interfere with the fluorescence.
