Application

Spermidine has been used in the preparation of gold beads[1] and as a component in nuclei isolation buffer (NIB).[2]
Spermidine is suitable for use in molecular biology buffers and for precipitation of DNA >100 bp in length. It was used to deliver DNA in plants and mammalian cells by particle bombardment technique.[3][4]

Biochem/physiol Actions

Inhibits neuronal nitric oxide synthase (nNOS). Binds and precipitates DNA; Stimulates T4 polynucleotide kinase activity.
Inhibits neuronal nitric oxide synthase (nNOS). Binds and precipitates DNA; may be used for purification of DNA binding proteins. Stimulates T4 polynucleotide kinase activity.
Spermidine helps to maintain the proper activity of gene expression.[5] This polyamine plays a key role in intermediate metabolism. It also serves as an inhibitor of the acetyl transferase activity of E1A-associated protein p300 (EP300).[6] Spermidine participates in cell growth.[7]

Disclaimer

Solutions should be sterile-filtered, not autoclaved, if sterile solution is necessary. Spermidine deaminates with time; solutions should be stored frozen. Prepare new solutions frequently.

General description

Spermidine belongs to the natural polyamines, that are found in chloroplasts and in bacterial or eukaryotic cells.[5]

Questions

1–7 of 7 Questions
  1. Can the reconstituted S0266 solution, which has been sitting at room temperature for 3 months, still be used in CUT&Tag assays to stabilize nuclei? Also, how can the integrity of spermidine be tested? If there's no more spermidine available to test the solution, should more be purchased?

    1 answer
    1. The integrity of Spermidine can be verified by Gas chromatography. The following publication may be of interest for protocol reference on Gas chromatography analysis of Spermidine.
      1. J Mass Spectrom. 2009 Aug;44(8):1203-10. doi: 10.1002/jms.1597.

      Helpful?

  2. What is the solvent that spermidine is dissolved in?

    1 answer
    1. This product is a neat or pure liquid and has not been diluted in a solvent. However, to further dilute this product, it is recommended to use water at 50 mg/mL water.

      Helpful?

  3. How to store S0266-5G? which comes as liquid and what is the molaritty of it? which solvent to use to dilute it?

    1 answer
    1. This product can be stored in room temperature in its original format. The concentration of this product can be determined using the lot specific purity reported on the Certificate of Analysis, the density, and the compound molecular weight. This product has a density of 0.925 g/mL and a molecular weight of 145.25 g/mol. The minimum purity is 98%, so the molarity will vary slightly from lot to lot. The molarity can range from 6.2M (98%) to 6.4M (100%).
      Molarity=Density x 1000 x Purity/MW

This product is supplied as a liquid or solid. A stock solution can be made by dissolving 1.45g in 10mL of water followed by sterile filtration with a 0.22um filter.

      Helpful?

  4. How to store S0266-1G which comes as liquid? alliquoted and store at -20C? How long it can be stored at this condition?

    1 answer
    1. Solutions should be stored as single-use aliquots at -20 ºC for no more than one month.

      Helpful?

  5. How frequently should new solutions be made?

    1 answer
    1. A stock solution can be made by dissolving 1.45g in 10mL of water followed by sterile filtration with a 0.22um filter. Solutions should be stored as single-use aliquots at -20 ºC for no more than one month.

      Helpful?

  6. Is this a spermidine solution or a neat liquid, and what is the concentration?

    1 answer
    1. Based on a density of 0.925 g/mL and a molecular weight of 145.25, this neat liquid has an effective molarity of 6.4 M.
