Description générale
La sérum albumine bovine (BSA) est une protéine dont la principale fonction biologique est la régulation de la pression oncotique du sang. La BSA est utilisée dans une grande diversité d'études et d'applications. Constituée de 583 acides aminés[1][2], la BSA est une protéine soluble dans l'eau qui présente un poids moléculaire théorique de 66 430 Da. Ses trois domaines homologues sont formés par six hélices α. Selon le pH, elle peut subir une isomérisation conformationnelle réversible. Lors du chauffage, sa structure native devient flexible et réactive.[3]
Application
La sérum albumine bovine (BSA) est utilisée dans un grand nombre d'applications, notamment :
- La BSA a été utilisée en culture cellulaire, par exemple comme supplément dans des milieux préparés pour la culture d'oligodendrocytes.[4]
- La BSA a été utilisée pour la dilution d'une solution d'anticorps primaires dans un protocole d'immunohistochimie.[5]
- La BSA a été utilisée comme étalon dans la méthode de quantification des protéines de Bradford.[6]
- La BSA a été utilisée dans un tampon de blocage et de perméabilisation pour des analyses par immunofluorescence.[7]
Actions biochimiques/physiologiques
Certains épitopes conformationnels et certains épitopes de la séquence primaire de l′ASB sont soupçonnés d′être des allergènes impliqués dans les allergies humaines au bœuf et au lait.
Notes préparatoires
L'albumine sérique est parfois appelée Fraction V. Cette dénomination provient de la méthode originelle de Cohn consistant à fractionner les protéines sériques par précipitation dans de l'alcool froid. L'albumine sérique a été trouvée dans la cinquième fraction d'éthanol selon la méthode de Cohn. Depuis, l'expression "Fraction V" est utilisée par certains pour décrire l'albumine sérique, quelle que soit la méthode de préparation employée. D'autres utilisent cette appellation pour décrire de l'albumine sérique purifiée par des méthodes de fractionnement à l'éthanol qui ont beaucoup évolué depuis le développement de la méthode de Cohn d'origine. Sigma-Aldrich fabrique et distribue des albumines sériques purifiées à l'aide de diverses méthodes primaires, notamment la véritable méthode de fractionnement de Cohn, des méthodes de fractionnement à l'éthanol modifiées, le choc thermique et la chromatographie. Les étapes de purification supplémentaires peuvent inclure une cristallisation ou une filtration sur charbon.
