Application

Creatine (Cr) and phosphocreatine (PCr) are involved with rapid ATP production primarily in skeletal muscle tissue via the action of creatine kinase(s). Creatine may be used as a supplement to study its uptake mechanism and metabolism of action.

Biochem/physiol Actions

Creatine is a nitrogenous compound that acts as a high-energy reservoir for the rapid regeneration of ATP.
Creatine is a nitrogenous compound that acts as a high-energy reservoir for the rapid regeneration of ATP. Approximately 95% of creatine is found in skeletal muscle, primarily as phosphocreatine. Creatine can be acquired through dietary consumption or formed from L-arginine, glycine, and L-methionine in a multi-step reaction that occurs in the kidneys and liver. Creatine is then transported to muscle tissue. Creatine supplementation is used for the enhancement of sports performance, primarily by increasing muscle mass. Creatine is also being investigated as a treatment of neuromuscular diseases, where it may aid in neuroprotection and by improving the cellular bioenergetic state.

Questions

1–4 of 4 Questions
  1. What is the appropriate diluent for this product? PBS or DMSO to treat the cells.

    1 answer
    1. The solubility of this product is tested at 50 mg/mL in water. The solubility of this material in DMSO has not been determined. PBS is expected to be a suitable solvent, however the rate may be somewhat decreased. Gentle heating may be required for complete dissolution.

  2. I am going to treat the hmc3 cells with Creatine. However, I am unable to find a concentration in the papers that used this product. Could you kindly help me with this?

    1 answer
    1. A validated concentration for HMC3 cells treated with this product was not found. Creatine anhydrous is suitable for mammalian cell culture. In vitro studies using creatine report concentrations in the millimolar range. Recommend a stepwise dose–response: 0.5, 1, 2.5, 5, and 10 mM, with 5 mM as a practical starting point, accompanied by cell viability assessment.

  3. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  4. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf
