General description

Lipids are fundamental building blocks of cells and play important cellular roles. Lipids are key components of the plasma membrane and other cellular compartments, including the nuclear membrane, endoplasmic reticulum, Golgi apparatus, and trafficking vesicles such as endosomes and lysosomes. Intracellular lipid droplets are cytoplasmic organelles involved in the storage and regulation of triglycerides and cholesterol esters. Lipids have been implicated in various diseases including heart, vascular and neurological diseases.

BioTracker Lipid dyes rapidly stain lipid droplets in live or fixed cells with no wash step and minimal background staining of cellular membranes or other organelles. Cells also can be fixed and permeabilized after staining.

The BioTracker 488 Green Lipid Dye has an excitation around 430 nm, and can be excited equally well at 405 nm or 488 nm. In cells, it stains lipid droplets with bright green fluorescence using the FITC detection channel.

Questions

1–3 of 3 Questions
  1. Good afternoon, I would like to to know if it is possible to use the reagent also by flow cytometry application, and if there are any references or examples of it having been successfully used in this context, thank you

    1 answer
    1. This product is recommended for use in fluorescence microscopy and has not been evaluated for use in Flow Cytometry. However, the following citation reports the use of BioTracker 488 Green Lipid Droplet Dye in Flow Cytometry.

      https://pmc.ncbi.nlm.nih.gov/articles/PMC11061182/

      Helpful?

  2. Are there any solutions for the high background issue experienced when using the product with heart tissue cells?

    1 answer
    1. The high background issue could be due to the use of antifade, which is not recommended. Instead, it's suggested to use PBS as the mounting medium and nail polish as the coverslip sealant. Additionally, using other mounting media such as EverBrite™ or FluoroShield™ may impact staining patterns, so it's important to consider the recommended mounting methods to avoid issues with signal intensity and staining patterns.

      Helpful?

  3. Is BioTracker 488 Green Lipid Dye (SCT144) toxic for the cells? Can they be monitored for extra time after staining? How long does the stain last in this case?

    1 answer
    1. No cytotoxicity of this dye has been observed with incubation periods of up to 24 hours. Cytotoxicity beyond the 24-hour incubation period has not been studied. Suitability for extended incubation times would have to be determined by the end user.
