General description

Amicon® Ultra-2 centrifugal filter devices offer fast ultrafiltration. It has the potential to recover high concentration factors and easy concentrate from dilute and complex sample matrices. Amicon® Ultra-2 devices are supplied non-sterile and are for single use only.

Application

  • Ultrafiltration with 10,000 Dalton molecular weight cutoff Ultracel® membrane.
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids, or microorganisms.
  • Purification of macromolecular components found in tissue culture extracts or cell lysates.
  • Primer removal, PCR cleanup, removal of linkers or macromolecular labels from a reaction mix, and protein removal prior to HPLC.
  • Desalting, buffer exchange and protein dialysis.
  • Size exclusion and protein fractionation.
Amicon® Ultra-2 Centrifugal Filter Unit has been used:
  • to reduce large volume biofluids prior to extracellular vesicle (EV) isolation or to concentrate EVs[1]
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids (DNA/RNA samples, either single- or double-stranded), microorganisms, column eluates, and purified samples
  • Purification of macromolecular components found in tissue culture extracts and cell lysates, removal of primer, linkers, or molecular labels from a reaction mix, and protein removal before high-performance liquid chromatography (HPLC)
  • Desalting, buffer exchange[2], or diafiltration

  1. At what speed should the Amicon® Ultra Centrifugal Filter, 10 kDa MWCO UFC201024 be used? Besides, what is the maximun g-force supported by the Amicon?

    1 answer
    1. The speed needs to be evaluated. However, we know that the maximum speed should never be exceeded. It is best to run below the maximum speed to not risk breakage in case the centrifuge has not been calibrated recently. Here are our specifications for the Amicon Ultra 0.2: Maximum relative centrifugal force Swinging bucket rotor 4,000 × g for concentration spin, 1,000 × g for recovery spin Fixed angle rotor 7,500 × g for concentration spin, 1,000 × g for recovery spin
      NOTE: When spinning viscous solutions such as undiluted serum or plasma, do not exceed 5,400 x g. Furthermore, if you are trying to concentrate proteins, often, proteins do not like to be spun at a high speed or they can precipitate. Therefore, let's say your centrifuge is run at 2000 x g. This should work fine. It may take longer or it may not. This depends on initial and final desired volume when stopped.

  2. Can Amicon® Ultra Centrifugal Filter, 10 kDa MWCO UFC201024 to be in the Centrifuge as 4 mL Centrifugal Filter without adaptors?We have adaptors for 15 mL TUBES

    1. Amicon Ultra-2 devices should be used in a centrifuge with swinging bucket or fixed angle rotor with wells/carriers that can accommodate 17 mm × 100 mm tubes. This is the same well/carrier size that is recommended for the Amicon® Ultra-4 devices. To avoid damage to the device during centrifugation, make sure it is properly assembled and seated at the bottom of the rotor. The rim of the concentrate collection tube should be inside the rotor well. Check clearance before spinning.

  3. Hello. Does these 2 ml 10 kDa filters fit into the same centrifuge rotor as the 0.5 ml 10 kDa amicon filters?

    1. No, the Amicon-0.5 devices require a microfuge, while the Amicon-2 devices require a centrifuge with adapters for 5 ml tubes.
