Analysis Note

The following is a list of properties associated with our agaroses:
Sulfate content - used as an indicator of purity, since sulfate is the major ionic group present.
Gel strength - the force that must be applied to a gel to cause it to fracture.
Gel point - the temperature at which an aqueous agarose solution forms a gel as it cools. Agarose solutions exhibit hysteresis in the liquid-to-gel transition - that is, their gel point is not the same as their melting temperature.
Electroendosmosis (EEO) - a movement of liquid through the gel. Anionic groups in an agarose gel are affixed to the matrix and cannot move, but dissociable counter cations can migrate toward the cathode in the matrix, giving rise to EEO. Since electrophoretic movement of biopolymers is usually toward the anode, EEO can disrupt separations because of internal convection.

Application

Agarose has been used in the preparation of microgel slides employed in lysis and electrophoresis.[1] It has also been used in the gel electrophoresis to separate restriction enzyme digested products in catechol-O-methyltransferase (COMT) genotyping[2]
Ideal for the separation of small DNA fragments (e.g., PCR products) differing in size by as little as 2% and compares to the resolution of DNA in polyacrylamide gels. High resolution agarose is also ideal for resolving amplification fragment length polymorphisms (AmpFLP), short tandem repeats (STR) and tri- and tetranucleotide repeats.

General description

Agarose is a polymer extracted from agar or agar-bearing marine algae. This purified linear galactan hydrocolloid comprises alternating co-polymers D-galactose and 3,6-anhydro-L-galactose units connected by α-(1→3) and β-(1→4) glycosidic bonds.[3] Agarose is highly biocompatible and possesses variable mechanical and diffusion properties.[4] Agarose can be used as a gelling agent, to separate nucleic acids electrophoretically.[5]

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. Is this agarose high-resolution, and suitable for PCR applications?

    1. The High Resolution Agarose, product number A4718, is an intermediate-melting agarose that offers approximately double the resolution capability compared to standard agarose. It is capable of resolving PCR products and DNA fragments ranging from 200-800 bp that differ in size by as little as 2%. This makes it ideal for separating small DNA fragments, such as PCR products, with very small size differences, offering a resolution comparable to that achieved with polyacrylamide gels.
