General description

Amicon® Ultra 15 mL centrifugal devices with Ultracel® 100,000 Dalton molecular weight cutoff filters provide fast ultrafiltration, and reliable retention of macromolecules. For example, IgG (156 kDa) at 1 mg/ml initial concentration is retained at >90% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.

The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon® Ultra 15 mL filters deliver the highest recovery with short spin times.

Application

  • Ultrafiltration with 100,000 Dalton molecular weight cutoff Ultracel® membrane.
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids, or microorganisms.

  1. I see the device is only compatible less than 100 mM imidazole. However the his-tagged protein usually washed higher than 100 mM imidazole. what will happen if it is used to concentrate sample with imidazole higher than 100 mM?

    1. At the time the User Guide was written, 100 mM was the highest concentration tested. Subsequent testing has been conducted with concentrations as high as 300 mM. Higher concentrations of imidazole are not recommended for concentration. Using imidazole concentrations above 100 mM is not expected to damage the device; however, high salt solutions, including imidazole, may lead to sub-optimal results. Sub-optimal results can include slower flow rates, poorer recovery, and increased detection background or interference in model systems.

  2. Can it be fit into 50ml centifuge rotor

    1. Yes, Amicon Ultra-15 units are compatible with adapters for 50 ml tubes.

  3. Filters with smaller (MWCO) values (3K/10K) tend to produce a higher filtrate volume in a shorter time compared to those with larger MWCOs (e.g., 100K).Why do filters with smaller MWCOs appear to elute faster, even though they have smaller pore sizes?

    1. Each liquid being concentrated in centrifugal devices, each type of substance in the liquid, each amount of particles in a liquid, and each initial volume, type of size, and surface area of a device is unique. Centrifuges are spun at a specific speed based on which type of centrifuge is being used. Therefore, there are many variables involved in the reduction of volume from one initial volume to another, and thus, the end result of the time it takes to reduce volume is different. We as a vendor cannot comment on a customer's specific observation and final result explaining volume reduction and time of a sample.

  4. The product comes with the tube and filter as one piece. Is this correct? how can i get under 100kDa liqud?

    1. No, the filter component is removable to facilitate sample recovery.

  5. Temperature limit?

    1. The Amicon Ultra devices are rated for Room Temperature environments and "normal laboratory conditions". We have not studied a set limit but do know that if stored in a warehouse at higher temperatures and humidities, the devices can be affected.

  6. Is it reusable? How many times I can fill it up again? Amicon Ultra 15. First time I fill it up with 15 ml, and after can I use it again for other 15 ml....and again?

    1. The Amicon Ultra 15 is designed for single use but can be used for repeated spins with the same sample. However, repeated use will cause clogging of the membrane, leading to slower flow rates and longer spin times.

  7. Hello, Are these tubes valid for AAV concentration? And if so, are these valid for all serotypes? Also, the user guide says that the membrane's material is 'Ultracel® low binding regenerated cellulose'. What's the difference with the PES membrane?

    1. These devices can be used for virus concentration but are not validated for any specific type. Both PES and regenerated cellulose membranes have low binding, but PES has a faster flow rate.   8. Hi, I have plan to use the Amicon Ultra-15 100K for Exosome extraction from cell culture supernatant, could you please let me know what would be the best speed (rpm/rcf) and time for doing that? Thanks

    1. Typically, Amicon-15 devices are spun at 4,000 x g (relative centrifugal force) for exosome isolation. However, spin times may need to be optimized, based on the sample type and volume.   9. What is the minimum starting volume I can load on Ultra 100K (100,000 MWCO) devices? Can I load as little as 500 µL?

    1. This product is intended for use with 4-15 mL initial sample volumes. The device has not been evaluated for use with volumes smaller than those recommended. While it may be possible to concentrate samples as low as 500 uL, the recovery volume cannot be predicted.   10. Does the Amicon Ultra has residues that may released into the concentrated product', like Beta-Glucan?

    1. These devices contain trace amounts of glycerine. If this material interferes with downstream analysis, rinse the device with buffer or Milli-Q® water prior to use. Should interference continue, rinse with 0.1 N NaOH followed by a second spin of buffer or Milli-Q® water. NOTE: Do not allow the membrane to dry out following rinse steps. If the product will not be used immediately after rinsing, leave the final buffer or water on the membrane until the usage. Please see the link below to review the full product User Guide:
