Szczepy gospodarza RosettaBlue są pochodnymi NovaBlue, które łączą wysoką wydajność transformacji i mutacje recA^endAlacIq ze zwiększoną ekspresją białek eukariotycznych, które zawierają kodony rzadko używane w E. coli. Szczepy te dostarczają tRNA dla AGG, AGA, AUA, CUA, CCC i GGA na plazmidzie odpornym na chloramfenikol. Geny tRNA są napędzane przez ich natywne promotory. Niebiesko-białe badania przesiewowe nie są możliwe w przypadku szczepów RosettaBlue(DE3) ze względu na obecność sekwencji kodującej peptyd α lacZ w profagu λDE3.

DE3 wskazuje, że gospodarz jest lizogenem λDE3, a zatem posiada chromosomalną kopię genu polimerazy RNA T7 pod kontrolą promotora lacUV5. Takie szczepy są odpowiednie do produkcji białek z genów docelowych sklonowanych w wektorach pET poprzez indukcję IPTG.

Szczepy pLysS wyrażają lizozym T7, który dodatkowo tłumi podstawową ekspresję polimerazy RNA T7 przed indukcją, stabilizując w ten sposób rekombinanty pET kodujące białka docelowe, które wpływają na wzrost i żywotność komórek.
