Opis

General description

Human phosphatase-substrate networks are mapped based on the literature and protein-protein interaction (PPI) database. Based on gene expression data, the possible network of phosphatase-substrate-kinase is predicted.

Application

The substrate has been used to determine the alkaline phosphatase (ALP) activity in human osteoblast-like Saos-2 cell lysates. ALP acts as an early marker of osteoblast differentiation that is expressed just before matrix mineralization begins.

Chromogenic substrate for use with alkaline and acid phosphatase ELISA procedures. Produces a soluble end product that is yellow in color and can be read spectrophotometrically at 405 nm. The pNPP reaction may be stopped with 3 N NaOH and read at 405 nm.

Preparation Note

Contains 40 mg substrate per tablet

Dissolve tablets to the desired concentration in either of the following buffers: 0.1 M glycine, pH 10.4, with 1 mM MgCl₂ and 1 mM ZnCl₂ or 1 M diethanolamine, pH 9.8, with 0.5 mM MgCl₂. Typically, a pNPP concentration of 1 mg/mL is used.
