Analysis Note

The NIH assay procedure uses 0.2 ml diluted plasma (1:1 with saline) as a substrate and 0.1ml of thrombin sample (stabilized in a 1% buffered albumin solution) based on a modification of the method of Biggs. Only clotting times in the range of 15-25 seconds are used for determining thrombin concentrations.

Application

Thrombin is used for site specific cleavage of recombinant fusion proteins containing an accessible thrombin recognition site for removal of affinity tags. Thrombin has been used in a study that compared efficacy and safety of low-molecular-weight heparins (LMWHs) to other anticoagulants as venous thromboembolism prophylaxis in major orthopedic surgery. [1]

Biochem/physiol Actions

Serine protease that selectively cleaves Arg-Gly bonds in fibrinogen to form fibrin and fibrinopeptides A and B.

  1. What is the stability of this material after Lyophilization? Can I save it in the -20^0C?

    1. This product is provided as a lyophilized powder. When stored appropriately at -20ºC, the material is quite stable and not assigned an expiration or retest date. A warranty period of one year from the date of shipment applies. Following reconstitution, stock solutions remain active for one week at 2 – 8ºC. Solutions are most stable at pH 6.5, as a pH >7 will greatly reduce thrombin activity. Since thrombin solutions adsorb to glass, it is recommended to aliquot solutions in plastic tubes and store at –20 ºC for long-term storage. Various literature sources indicate solutions may be most stable when stored at -70ºC. The long term stability of this product in solution has not been established.
