Application

Duolink®proximity ligation assay(PLA®) allows for endogenous detection of protein interactions, post translational modifications, and protein expression levels at the single molecule level in fixed cells and tissue samples.

To perform a complete Duolink® PLA in situ experiment you will need two primary antibodies (PLA, IHC, ICC or IF validated) that recognize two target epitopes.This starter kit supplies all other necessary reagents for 30 Duolink® PLA reactions, which include a pair of PLA probes (Anti-Rabbit PLUS and Anti-Mouse MINUS), red detection reagents, wash buffers, and mounting medium. Note that the primary antibodies must come from the same species as the Duolink® PLA probes. Analysis is carried out using standard immunofluorescence assay equipment.
Specificity
The Duolink® In Situ Red Starter Kit Mouse/Rabbit requires one primary antibody from mouse and one primary antibody from rabbit. Red fluorescence detection reagents are often used with Texas Red® filter.

Application Note
Two primary antibodies raised in different species are needed. Test your primary antibodies (IgG-class, mono- or polyclonal) in a standard immunofluorescence (IF), immunohistochemistry (IHC) or immunocytochemistry (ICC) assay to determine the optimal fixation, blocking, and titer conditions. Duolink® PLA in situ reagents are suitable for use on fixed cells, cytospin cells, cells grown on slide, formalin-fixed, paraffin embedded (FFPE), or tissue (fresh or frozen). No minimum number of cells is required.

      This kit contains the following components:

      Duolink® In Situ PLA® Probe Anti-Rabbit PLUS, Affinity purified Donkey anti-Rabbit IgG (H+L) (DUO92002)
      Duolink® In Situ PLA® Probe Anti-Mouse MINUS, Affinity purified Donkey anti-Mouse IgG (H+L) (DUO92004)

      These components are each comprised of the following:
      • 5x PLA Probe Anti-Rabbit PLUS - Donkey anti-rabbit secondary antibody conjugated to oligonucleotide PLUS
      • 1x Blocking Solution - Reagent for blocking of the sample
      • 1x Antibody Diluent - For dilution of PLA probes and primary antibodies

      Duolink® In Situ Detection Reagents Red (DUO92008)

      This component is comprised of the following:
      • 5x Ligation - Contains oligonucleotides that hybridize to the PLA probes and all components needed for ligation except the Ligase
      • 1x Ligase (1 unit/?L)
      • 1x Polymerase (10 units/?L)
      • 5x Amplification Red - Contains all components needed for Rolling Circle Amplification (RCA) except the Polymerase. It also contains oligonucleotide probes labeled with a fluorophore that hybridize to the RCA product.

      Duolink® In Situ Wash Buffers, Fluorescence (DUO82049)
      Wash Buffer A is used after the PLA Probe incubation step and Wash Buffer B is used after incubation with the amplification reagents.

      Duolink® In Situ Mounting Medium with DAPI (DUO82040)
      Duolink® In Situ Mounting Medium with DAPI is ideal for nuclear staining and preserving signals generated with the Duolink® In Situ Detection Reagents for fluorescence microscopy. Note: Counterstaining with Cy®2 is not recommended.

      The Duolink® In Situ Microplate Nuclear Stain, Anti-Fade is sold separately.

  2. Which fixation method (formalin, PFA, ethanol, aceton/methanol) do you recommend to use for fresh frozen tissue slides before applying the PLA?

    1. The choice of fixation techniques varies between sample types. Academic literature should be consulted to determine the proven methods of fixation for the specific frozen tissue types used in IHC and PLA experiments to best address this question.

  3. Does this kit contain antibody diluent and blocking solution? Or do you have to buy them separately?

    1. The kit contains antibody 1X diluent and 1X blocking solution, which can be found in the component DUO92002 and DUO92004.

  4. Hello, Is this kit also suitable for flow cytometry-based analysis of the data?

  5. Can the Duolink kits be used with a Mouse IgM antibody?

    1. Duolink kits may be compatible with a Mouse IgM antibody, provided that the antibody contains the light chain. The Anti-Mouse MINUS probe specifically interacts with mouse IgG (H+L) and the light chains associated with IgM. Therefore, if the Mouse IgM antibody includes the light chain, it should be suitable for use with the Duolink kits.

  7. I want to study the interaction of two cell surface proteins using PLA kit . My primaries are mouse and rabbit. Does the kit have the two secondary and also all the components that i would require to carry out the assay. Can you advice?
